Flow cytometry is known as cell "CT", and its application fields include hundreds of applications in clinical diagnosis, cell biology research, environmental monitoring, biopharmaceutical and so on. The flow cytometer can rapidly measure, store and display a series of important biophysical and biochemical characteristic parameters of dispersed cells suspended in liquid, and can sort out the specified cell subpopulation according to the preselected parameter range. Foreign related companies occupy the domestic market share of up to 71%. Limited by the core components and card neck technology, the technology and commercial products of high-end flow cytometer have been mastered by foreign companies, which greatly limits the development of technology and industry in China's scientific research and analytical instrument industry.
In view of the current situation that the flow cytometer sorter of high-end life science instruments is monopolized by foreign products, the Wang Ce team of Suzhou Medical Institute broke through the key core "stuck neck" technology of 30-color flow analysis and high-throughput sorting on the basis of the previous three-laser and ten-color flow cytometer. A real-time determination speed of cells of 80000 cells/sec under 32 gates has been achieved. The self-developed fluid cell can achieve sample focus flow position <5μm and high-speed droplet generation, with a minimum nozzle aperture of 70μm. The high-end flow cytometer with independent intellectual property rights has been developed, which has broken the relevant technical monopoly and reached the international advanced level. The instrument has passed electromagnetic compatibility, safety regulations and environmental adaptability testing, and its technical readiness has reached level 8. Relevant achievements authorized more than 20 invention patents.
The instrument has been applied and demonstrated on the flow platform of the Institute of Biophysics, Chinese Academy of Sciences and Changchun Institute of Applied Chemistry, Chinese Academy of Sciences, and has been well received, providing a domestic weapon for life science research in China. This work was supported by the "High Performance Flow Cytometer Sorter" project of the national key research and development plan. The project leader is researcher Ma Yuting (member of the Youth Promotion Association of the Chinese Academy of Sciences).
